dc.description.abstract | La distrofa miotónica tipo 1 (la enfermedad muscular hereditaria mas común en el adulto joven) es causada por una expansión de la tripleta citosina-timinaguanina (CTG), la cual es inestable en las líneas germinal y somática. Aunque la fsiopatología de esta enfermedad no se conoce todavía, existen tres hipótesis para explicarla: la haploinsufciencia, la expresión alterada de genes vecinos y la ganancia de función del ácido ribonucleico (ARN). Ésta última hipótesis ha tomado fuerza recientemente y consiste en que el ARN con la expansión es retenido en el núcleo, donde forma agregados ribonucleares, que alteran la función de las proteínas involucradas en el procesamiento alternativo del ARN. Con la función proteica alterada, se presentan en el paciente las isoformas fetales de proteínas importantes para la función muscular y cerebral normal. Por esta razón, los agregados ribonucleares son posibles blancos terapéuticos, e incluyen el ARN con la expansión, el complejo ARN/proteínas del procesamiento del ARN o solo las proteínas del procesamiento del ARN. Algunas de las estrategias enfocadas en posibles terapéuticas incluyen los oligonucelótidos antisentido, los ARN deinterferencia y las moléculas pequeñas; todas ellas buscan la liberación de las proteínas unidas al ARN que tengan la mutación, con el fn de que lleven a cabo su función celular normal. Varias de estas estrategias han logrado gran éxito, pues han podido restablecer los patrones de procesamiento alternativo de los individuos sanos, el fenotipo muscular normal y contrarrestar algunos de los síntomas de esta enfermedad. Con estas técnicas, a deferencia de hace cinco años, se puede vislumbrar ahora una esperanza en el horizonte, para un tratamiento más eficaz y efectivo contra la distrofa miotónica tipo 1. | es_ES |
dc.description.abstract | Type 1 myotonic dystrophy (the most common form of hereditary muscular disease in the young adult) is caused by the expansion of the cytosine-thymine-guanine (CTG) triplet, which is unstable in the germ and somatic lines. Although the pathophysiology of this disease is unknown, there are three hypotheses to explain it: a) haploinsuffciency, b) altered expression of neighbor genes and c) ribonucleic acid (RNA) gain of function. This latter hypothesis has gained followers and refers to a nuclear retention of the RNA containing the expansion, then RNA forms “foci” and alters the function of proteins involved in RNA alternative splicing. The altered protein function leads in the patient to the presence of fetal isoforms of important proteins for normal muscle and brain function. Thus, the “foci” become possible therapeutic targets and include the expanded-RNA, the splicing proteins/ expanded-ARN or just the splicing proteins. The strategies used to treat the disease include the antisense oligonucleotides, the interference RNA and small molecules, all of them seeking to release the proteins, which are bound to the expanded RNA, so that the proteins become able to carry out their normal function.Some of these strategies have succeeded by restoring the alternative splicing patterns seen in healthy individuals, the normal muscular phenotype and counteract some of the disease symptoms. These strategies, unlike 5 years ago, allow us to see a light in the horizon for a more effcient and effective treatment for type 1 myotonic dystrophy. | es_ES |
